Effects of Sucrose Concentration and Days after Anthesis on in Vitro Pollen Germination of Various Allium hirtifolium Ecotypes

Abstract
In vitro pollen germination represents a promising approach to accelerating genetic improvement in plant breeding. To date, there has been no research focused on the pollen germination of Allium hirtifolium. This study aimed to investigate the effects of various sucrose concentrations and days after anthesis on pollen viability and in vitro pollen germination rates in different Allium hirtifolium ecotypes. Six populations of Allium hirtifolium (Persian shallot) from diverse regions of Iran were assessed for pollen germination in vitro. Germination was conducted in a basal medium supplemented with sucrose at concentrations of 1, 2, 3, 4, and 5%. Pollen germination rates were recorded at 1, 3, 5, and 7 days post-anthesis. Additionally, pollen grains were stored at deep-freezing temperatures for 30 days. The results revealed that both sucrose concentration and ecotype significantly influenced pollen germination, with the most pronounced effect occurring in response to the 2% sucrose concentration. The optimal germination time post-anthesis was 1 day. Notably, no pollen germination was observed after 30 days of deep freezing. Several days after anthesis, sucrose concentration and ecotype significantly correlated with pollen germination. This study provided empirical evidence for the selection of optimal media, timing, and ecotypes considering the pollen germination of Allium hirtifolium when using in vitro conditions.
