Potential of Pre-treatment in Enhancing of Transformation Efficiency In-planta Agrobacterium-mediated Genetic Transformation on Soybean (Glycine max L.) Seeds

Abstract
The development of soybean through genetic modification continues to be carried out to meet market and consumer needs. However, the transformation efficacy was quite inconsistent. To improve this efficacy, various pre-treatment methods could be performed. Therefore, this study aimed to investigate the potential of soaking method as pre-treatment to elevate the in-planta transformation efficiency of soybean seed. This study was carried out by using soybean seed of Dega I variety. This research consisted of 4 infection methods. The first and second methods, soybean seeds were infected directly by soaking them in bacterial suspension for 30 and 60 minutes, respectively. The third and fourth methods, soybean seeds were soaked for 10 minutes as pre-treated in warm water for 30 and 45°C, respectively. After that, the pre-treated seeds were soaked in the bacterial suspension for 30 minutes. Transformation efficiency was confirmed by detecting CaMV-35S promoter and nptII genes through PCR. Successful transformation was marked by the presence of 235 bp and 550 bp sized DNA bands, respectively. Results showed that the transformation efficiency was successfully increased up to 30% if the seed was soaked in warm water at 35°C and inoculated for 30 minutes. Compared to those pre-treated ones, the efficacy obtained from direct infection without soaking was ranging from 4 – 21%. This study highlighted that seed soaking could be a practical heat pre-treatment method to increase the efficacy of in-planta genetic transformation in soybean seed.
